Description
Myristoylated and Unmyristoylated-CA-recoverin structures differ significantly from one another, providing insight into the functioning the mystristoyl-CA switch. Making the assumption that the x-ray structure of one CA2+-unmyristoylated recoverin was a good approximation to the R form the myristoylated protein, Ames et al. compared the two proteins. It is found that the carboxy-terminal domains of the two structures are quite similar, while their amino-terminal domains are very different (2.2+/-0.1 A rmsd versus 6.9+/- 0.1 A). The two helices of EF-1 are nearly antiparallel in the CA2+-free protein, contrasting with their nearly perpendicular orientation (108 deg.) in the CA2+-bound protein. Mechanism seems to involve a backbond hinge rotation at the domain linker.
[June 1998 Update: The structure of Ca-bound myristoylated protein is indeed very much like that of the unmyristoylated Ca-bound protein, as anticipated (they are both in the R state). Per correspondance, M. Erickson]
